I really don’t have a set method. It’s mostly intuitive. I’ll usually start with a sample that has caught my ear and try to find a few more that fit the mood. Sometimes I use the sequencer, sometimes I don’t. There’s usually either Eventide Blackhole and/or Velvet Machine in the mix.

I usually use one-shot samples. Occasionally I will use a longer sample and chop bits out of it.
I’m not sure I know what you mean by “play the effects.”
By "play the effects" I believe @oscillotus means manipulating Koala's effects section while recording the output.
